Design Notes

Our team had to consider an optimal UAS - core promoter pair, and the study by Decoene et al. showed that cpTEF_6-I was the strongest synthetic promoter, and had the short length optimized protein expression. However, depending on its combination with the UAS, protein expression could be further modified with weaker promoter sequences. The UASfec should be used before the core promoter and a 5'UTR should precede the start codon of the insert of interest.

TATA-like sequence is GATTTAAA (2-9)
